Cold chain and handling

Freeze-dried peptides tolerate transit far better than solutions do. That tolerance is not unlimited, and it ends at reconstitution.

Dry material is the stable form

Water participates in most of the reactions that degrade peptides. Removing it by freeze-drying slows hydrolysis, deamidation and several other routes to the point where a well-dried peptide stored cold and dark has a usable life measured in years rather than weeks.

This is why material ships as a dry solid. A short period at ambient temperature during transit is a normal and expected part of the process for lyophilized material.

Reconstitution changes the arithmetic

Once a peptide is in solution the protective effect of dryness is gone, and stability is then governed by temperature, pH, the solvent used and how many times the vial is opened. Solutions are handled cold and used within a much shorter window than the dry material they came from.

Practical handling

  • Move material to its documented storage condition on arrival rather than leaving it at ambient temperature
  • Keep vials dark; several peptide side chains are photosensitive
  • Avoid repeated freeze-thaw cycling, which is more damaging than continuous storage at a single temperature
  • Allow a cold vial to reach room temperature before opening, so that atmospheric moisture does not condense onto the cold solid
