Copper peptide chemistry

GHK-Cu is a coordination complex. The copper is part of the molecule, not an additive.

How the complex forms

The GHK tripeptide binds a copper(II) ion through nitrogen and oxygen donor atoms from its histidine imidazole, the terminal amino group and backbone carbonyls. The result is a stable square-planar complex with distinct properties from the free peptide.

Why it matters practically

The complex has its own solubility and stability behaviour. Solvents or conditions that would suit an uncomplexed tripeptide can destabilise the copper coordination, which is why reconstitution guidance for this compound differs from the general case.

Chelating agents and some buffer components compete for copper and can strip it from the peptide, leaving two components rather than one.

Why copper at all

Copper is a cofactor in several enzymes involved in connective tissue, including lysyl oxidase. Much of the research interest in the complex concerns copper delivery and the redox chemistry the metal makes available, neither of which the free peptide provides.
